I’m sure you’ve all heard about IBM. They’re one of the pioneers of the PC industry, but nowadays, virtually no one uses IBM computers. Yet, IBM is still one of the largest companies in the world with a market cap of over $100 billion, so how does IBM even make money? Well, throughout IBM’s history, they were almost always a B2B company as opposed to a consumer brand. This is quite ironic given that they’re most well known for their PCs in the 1980s, but this was actually the exception for IBM, not the norm. After the dot-com bubble burst, IBM sold off its PC division to Lenovo and they started focusing on their B2B business once again. This consisted of producing server racks, networks, databases, and setting up cloud infrastructure. All of these background functions allowed IBM to rise to prominence once again, but with the rise of competition within these markets, IBM is struggling once again. Their revenues have nearly halved within the past 10 years and it’s not clear if they can compete against the juggernauts like Amazon and Microsoft. Back when computers were very limited in color, and pixels are huge, my brothers I often play Round 42. 42 rounds of pixelated alien invaderesque fun! On our IBM PC-XT, with 4 color CGA monitor, most of the games are 4 colors: Black White, and either ugly red and green, or cyan and white. Somehow, this game got all of these colors! The game was hard, and the final enemy of each round moves ridiculously fast. Then we found that F2 fires the zapper weapon :P

A "spurt" sound isn't something you ever want to hear from your monitor... I knew I wasn't going to get away with leaving it shifting between blue and red for long, but I didn't think it would be this short. If I had seen the cap on the connector board for the CRT initially then I wouldn't have had to take the whole thing apart. But I kind of wanted to get an overview of what was going on inside anyway so I don't mind the extra work having been put in. But if yours blows a color cap you can easily avoid the most difficult part of removing the tube. I'm happy with how this turned out. I will be getting better capacitors later for it, but it isn't a rush. I think I made all the colors a bit dim on the adjustment on the back of the tube. That's probably why it seemed so much darker afterwards. Really, I should have done the calibrations at about 75% brightness. Oh well, it's not too difficult to get to that so I can always fix it later. I was never aware of this computer "back in the day" ... I remember the IBM Convertible PC coming out with it's neat pop-up twin 3.5" floppy drives, but hadn't realised about this earlier Portable Personal Computer. It uses an earlier version of the IBM PC/XT (Model 5160) motherboard ... oh, and yes, the keyboard uses an RJ11 connector and not RJ45, duh. Got this sometime in 2018 for quite a good price - picked it up in Sydney so no shipping charges either - bonus! Ironically, it was "around the corner" from where we used to live (for ten years) before leaving Sydney for Port. From my earlier research, it appears to have an AST "SixPackPlus" memory expansion and serial I/O card, a standard CGA card, a hard drive plus an unknown card. I originally upgraded the BIOS to the latest available but then got stuck trying to get a CF drive and/or FlashFloppy drive to work. I may have also installed an NEC V20 and/or 8087 math co-processor. Maybe. Now that I've got it out again, first thing is to check what's lurking in the power supply before doing anything else. Got the power supply back together, re-installed the cards I needed: CGA with composite out, AST SixPakPlus memory expansion, floppy controller and XT-IDE (rev. 4 from Glitchworks). Left the HDD controller out. And ... it worked! Well, it POSTs a "601" error which I assume is because there's no floppy drives connected. I was surprised ... managed to get to GW-BASIC via the XT-IDE ROM boot. After some jumper fiddling, I managed to boot off the CF card. I'm using a 1GB Compact Flash card ("Dick Smith Electronics" branded!) that I know worked on my Amstrad PC1640 (using a Lo-Tech XT-IDE card). So it booted into MS-DOS 6.22 and was working fine. I loaded up my favourite game of all time ... The Ancient Art of War. Man, I played that game for hours ... along with The Ancient Art of War at Sea ... even managed to recover some campaigns I created on 5.25" floppies from back in the 90s (with references to now-ex-girlfriends, for some reason). Next - try to get the floppy drives working. Not really necessary as I can just load stuff onto the CF card, and even re-use the original HDD front panel & activity LED for the CF card ... let's see. In this talk, I'll give an overview of the current status of support for IBM Z processors in both GCC and LLVM, touching on differences in available feature sets and performance results. In addition, I'll be talking about my experience working in the context of both compiler frameworks. Where did I find GCC easier to work with, and where does LLVM have its advantages? THE TOWER OF THIEVES A foul wind is blowing in the kingdom of Pentawa. It blows through towns and villages with gales of blood and tempests of destruction. It calls itself 'The Blue Wind'. 'The Blue Wind' is a group of ruthless thieves who are casting a shadow of fear over the countryside. They have taken over the ruins of an old castle abandoned long ago under mysterious circumstances; its strange, forboding atmosphere seems somehow fitting to their purposes. A man, wounded in battle and nearly dead from hunger and thirst, has wandered into Pentawa looking for assistance. He tells of a narrow escape from the castle, and of a beautiful princess and many brave warriors held captive there. He also speaks of a vile demon and a dragon imprisoned for ages who may soon be free again. The king has issued an order for all Sorcerian adventurers worthy of the name to respond to this man's plea for help. 'Go forth in the name of your ancestors and rid the land of this evil that calls itself 'The Blue Wind'. GOAL An outlaw band is using a deserted castle as a base from which to raid nearby villages. They have captured the princess of a neighboring country and her company of fighters, who came to the castle on business of their own. There may be a dragon imprisoned under the castle. Infiltrate the Tower of Thieves, and rescue the princess and her warriors from this band of robbers, who may be more powerful than they seem. ITEMS VASE: Located on the altar in the chapel of the deserted castle. PASS: Several outlaws carry passes signed by Garok that allow them to go restricted places in the castle. GARATEEN: A holy sword in a silver scabbard. The blade is silver carved with sacred runes. RED BALL & BLUE BALL: These objects have something to do with opening some of the doors in the castle. PENDANT: A gold necklace with a large gemstone hanging from it. It possesses a strong protective magic. MONSTERS GAR: A ferocious fish that inhabits the lake under the castle. It has sharp teeth and powerful jaws. MAJOR DEMON: A shapechanging demon that likes to take human form. Cruel and very deadly. SHADOW DRAGON: The dragon has been imprisoned in a dark narrow cave for many years. Its wings have degenerated so that it can no longer fly, but its ability to breathe fire has not been diminished.

THE CURSED SHIP From the ship's log of the Maria Regina: 17th day, month of Dagonn. Fine. A good west wind. Good progress. Toward sunset, the cabin boy found Jacob's body in the forward hold. His throat had been cut. There were no witnesses. I can think of no-one who might want to kill Jacob -- he was well-liked among the crew. He had been ill the last few days, but it seems unlikely that he committed suicide. 24th day, month of Dagonn. Rain. I am afraid for my son. I don't like the idea of his being here on the ship, now that I'm certain that someone aboard is a murderer. I've tried to warn him to be cautious, but he wouldn't listen. Maybe my timing was bad; his pet macaw just disappeared. We will be putting in at Pentawa soon, and I want him to stay with his mother. Tensions are running high on the ship. The sailors are beginning to whisper that we are cursed. 3rd day, month of Hydra. Calm. When we dropped anchor in Pentawa Harbor, some people came on board asking for passage. They are adventurers, it seems, off to right some wrong somewhere. They treat it like some kind of sport. I offered them free passage in exchange for convincing my son to stay ashore when we leave. They may find this task harder than battling dragons. GOAL The ship Maria Regina is anchored in Pentawa Harbor. One of the sialors has been brutally murdered, and the rest of the crew is anxious and uneasy. The Captain's son refuses to go ashore to the safety of his mother's house. If the captain's dilemmas can be solved, he might be willing to give you passage aboard the Maria Regina to exotic ports where adventure abounds. ITEMS HATCH KEY: A key of simple design, it unlocks one of the ship's cargo holds. GOLD JAR: On its surface is engraved in ancient letters, "the one that is hard to let go." BLOOD STAINED AXE: No murder weapon has yet been found. Could this be it? AUGER: A common shipboard tool, used to make holes in barrels. MACAW: The captain's son's beloved pet. It was startled by a ship's rat and flew away. The boy is very upset. CABIN KEY: Key to unlock one of the sailor's private cabins. DESK KEY: This key opens a drawer in a desk somewhere on the ship. MONSTERS RAT: The rats aboard the ship have become very aggressive lately. GHOST: Spirits who seek out the living to rob them of life energy. SEAGULL: The seagulls are beginning to exhibit strange behavior, including attacking people. OWL: These owls, like the other animals around here, have begun to attack people. OAKROT: At first glance, this monster resembles the oak trees from which it evolved. (Note: Oakrot is not actually in any version of the game!) ARCHDEMON: This demon takes the form of a huge devil-face. Burning rays shoot forth from its mouth.

If anybody can help me with model numbers that would be suitable for this project, that would be useful :o) 2) With the tape drive, I've also been informed that you can use generic tape drives of the era to talk to this kind of set up and don't specifically need an IBM system, can you advise me on this as I am a complete novice. 3) With the hard drive, I believe I need about 1.2gb of storage. Can I use 9332's in this set up (I've been told they may be troublesome), or do I need another kind of storage solution (please advise). 4) And now for the largest problem.....the OS. I know that I will need os/400 for this, but is there a way to get a copy without sourcing from ibm (not a rich guy so no way I can afford it from them). I've also been told that I would need a copy specific to my hardware configuration. Is this true, and is there any way around this? 5) Ok, the other thing is, does anybody know of anywhere I can get the hardware for this without paying premium prices? Heart of China, inspired by big-screen adventures like Raiders of the Lost Ark and High Road to China, was developed using the Dynamix Game Development System, the same framework used in Rise of the Dragon released a year prior. The artwork used a mixture of digitized photos of live actors and hand-painted backgrounds. With a tight production budget, Dynamix had to recruit the cast of actors from the company's own employees and their families. The game's fantastic atmospheric settings, employing great color palettes and beautiful hand-drawn backgrounds looked amazing at the time and still do. The narrative while a bit on the cheesy and stereotypical side still makes for a great adventure. The new IBM FlashSystem is a family of products meant to address the varying storage requirements of organizations ranging from startups to large-scale enterprises. As the company explains, different businesses have different needs for their storage, with things such as performance, scalability, data services, functionality and high availability just some of the considerations. Traditionally storage vendors have solved this the easy way, simply by providing a massive range of different storage products for different purposes. That’s all well and good, but as the number of storage platforms an organization uses rises, so does the level of complexity they need to deal with. Complexity is never a good thing when it comes to information technology. In the case of storage, it means using different management systems for each storage platform, different ways to troubleshoot problems, different application programming interfaces in order to interact with those systems, different paths to the cloud and many other headaches. Add to that, most enterprises use several systems from multiple providers, adding to the headaches they need to deal with. The complexity means that organizations often have to delay the deployment of new storage technologies, while it also limits their flexibility and drives up costs. In an exclusive interview with Wikibon analyst Dave Vellante on SiliconANGLE’s mobile livestreaming studio theCUBE, Ed Walsh, IBM’s general manager of Storage and Software Defined Infrastructure, said the storage landscape has evolved into the complicated mess it is today because in the past, there was really no other way of doing things. “There’s always been a difference between high-end solution sets and low-end solution sets,” Walsh said. “Everything was based upon the features people needed, for example right availability, the right price points, features, functions, availability. It made sense that you had to have a unique thing. But because of the innovation we’ve driven, you no longer have to do that.” Perhaps the single-most important innovation with the new IBM FlashSystem family is that each system runs the same software stack. IBM Spectrum Virtualize provides the same “enterprise class data services” for entry-level all the way up to its high-end systems, while also enabling “nondisruptive data movement” and cloud and container deployments with supported storage, the company said. FlashSystem also provides consistent APIs for automating the provisioning and management of storage, supporting approaches including bare metal, virtualized, containerized and hybrid/multicloud workloads. “Our platforms are now one: It’s going to be one software and one API to manage,” Walsh said. “We’re going to do simplification without compromise we’re gonna give you what you need. You’re going to need an entry level packaging, midrange and high-end, but it’s going to be one software allows you to meet every single price requirement and functionality.” The new storage family comprises the IBM FlashSystem 9200, a high-end system for the most demanding of enterprise storage requirements; IBM FlashSystem 9200R, which is for customers that need a system that’s fully assembled, installed and configured by IBM’s engineers; IBM FlashSystem 7200, which provides hybrid and multi-cloud functionality for midrange enterprise deployments; and a number of entry-level offerings, including FlashSystem 5010, 5030 and 5100. IBM further promises “six nines” availability across all price points, with two- and three- site replication, cross-site high-availability configurations and the option for guaranteed 100% data availability. FlashSystem is managed in the cloud with IBM Storage Insights, which provides artificial intelligence-based analytics for insights into best practice recommendations plus 24/7 access to support when it’s needed. AI also powers the IBM EasyTier capability that helps to make sure the right data is placed in the right tier at the right time. With it, the “hottest” or most-used blocks of data move up the tiers as necessary when they’re more frequently accessed, while “cooler” blocks of data move down. ...

I just picked up this IBM 5160 the other day and I'm really excited to try and get it going! I am stunned the HDD is still working in this computer! Who knows when it was last powered on, if the head was parked, or how roughly it's been treated. I'll probably make this computer my dual monitor system once I move a color card into it since its 8 ISA slots make it easier to waste one on a weird setup like that. I'm definitely leaving the original HDD in there though and am going to use it. It's just to cool to pull and replace with an XT-IDE. A Welcome Virus: Cracking the Viral Code for the Battle Against Cancer Chairman and founder of PsiVac, Prof. Ghassan Alusi, and the chief operating officer, Imad Mardini, discuss how the company’s proprietary oncolytic virus platform offers new hope for cancer patients. During a time when everyone actively fears viruses (especially THE virus) and their mutations, it is only cancer cells that have cause to worry about oncolytic viruses, and rightly so. These mutated viruses are administered directly into a tumor. Once inside, they crack open the tumor’s cells in a process known as lysing that provokes a strong response from the body’s immune system, which has, until then, ignored the cancerous cells. What’s more, the therapy’s attack doesn’t stop at a single tumor. The replicating and lysing viruses release previously hidden tumor-associated antigens (TAAs) that alert the immune system about cancer cells to attack all over the body. The body’s own immune system then goes on to destroy previously unrecognized tumors far removed from the initial injection point. The biotech company PsiVac advances this technology even further by creating a treatment platform that transforms the adenovirus, aka the common cold, into an especially powerful oncolytic virus. A precision modification in the virus’s DNA improves its efficiency against cancer cells while making it harmless to other cells, rendering the treatment at once more effective and safer for patients. “Now that the technologies of other forms of immunotherapy are gaining ground, and as cancer remains a major cause of mortality, we now understand there is a huge need for oncolytic viral therapy,” says Prof. Alusi, whose company has planned to start Phase 1 clinical trials later this year. Unlike other immunotherapies, such as patient-centric CAR T-cell therapy, oncolytic viruses can be made in relatively large quantities once their efficacy and safety have been proved. The release of TAAs means that the tumor is no longer successfully hiding from the immune system, and the body can begin to fight the disease by its own means. The cytotoxicity of a virus is the extent to which a virus attacks and destroys cells, often an undesirable event. However, with oncolytic viruses, this cytotoxicity works in a patient’s favor, thanks to gene editing, by being specifically designed to attack cancer cells. An agnostic oncolytic virus targets not only one or a group of cancers but is effective against all malignant solid palpable tumors. PsiVac’s modified adenovirus has proven agnostic so far, making it a powerful weapon in the fight against cancer.

The April 2020 publication in ACS Biomaterials, Science & Engineering illustrates just how fast bioprinting is moving, producing results that were unimaginable five years ago, facilitating the study of tissue models in basic science, disease modeling, and drug discovery. But Allevi is not stopping at Earth-bound breakthroughs. The US company has also secured funding for simultaneous bioprinting experiments on the International Space Station. Curious to Know More? Angiogenesis: From ancient Greek, literally the creation (genesis) of vessels (angio). In modern biology, the term refers to the formation of new blood vessels within a tissue. In bioprinting, the presence of angiogenesis means that the printed tissue is behaving and growing in a way that is akin to living tissue and is essential to the success of creating functional printed tissues and organs. Vascularization: Also related to the creation of blood vessels but, in contrast to angiogenesis, successful vascularization of a printed tissue is when an existing, printed structure of blood vessels is adopted by the tissue to delivering blood throughout the structure. Innervation: The process by which a tissue is supplied with nerves. In 3D bioprinting, supplying a printed tissue with nerves is the latest frontier of the technology as supplying tissue not only with blood but also nerves could possibly accelerate restoration of muscle function in vivo and create more complex tissue models for research on neurological diseases.

This approach combines the targeted delivery of the antibody with the cancer-killing power of the cytotoxic drug that would be too potent to be used on its own. A cytotoxic drug is a drug that contains a molecule toxic to cells, leading to cellular death. Used in traditional chemotherapy, these molecules attack both healthy and cancerous cells. When linked to an ADC antibody, they target only the tumor. Targeted delivery of a cytotoxin is when a cell-killing toxin is delivered to a specific type of cell, such as tumor cells. This specificity allows for effective cancer treatment with fewer unwanted side effects for the patient. Scaling-up production for bioconjugates involves moving from manufacturing small batches for clinical trials to large batches up to five kilograms for commercial production. This major challenge for companies is essential for the successful commercialization of ADCs.

Off-the-shelf Cell Therapy Remestemcel-L Targeting the Lethal Complication of the COVID-19 Pandemic Mesoblast CEO Dr. Silviu Itescu speaks to Lonza about the company’s advanced portfolio of anti-inflammatory allogeneic cellular medicines including remestemcel-L, which is currently being evaluated in a U.S. Phase 3 randomized controlled trial for acute respiratory distress syndrome (ARDS), the principal cause of mortality due to COVID-19 infection. Most COVID-19-related deaths result from ARDS, an overactive immune response that creates overwhelming inflammation in lung tissue. As the world suffers in the grip of this pandemic, some hope comes from Mesoblast. They have dedicated the past 15 years to developing a portfolio of product candidates based on mesenchymal lineage cells to treat severe inflammatory diseases such as graft versus host disease, advanced heart failure, and chronic low back pain. Their strategy for treating inflammatory diseases uses cellular medicines instead of small molecules or monoclonal antibodies, thereby treating an out-of-control immune system on a systemic level by impacting multiple inflammatory pathways at the same time. This positions them well to provide much-needed treatment for those COVID-19 ARDS patients left with few – or no – options. The Phase 3 trial was informed by the positive results from an emergency compassionate use protocol in patients on mechanical ventilation with COVID-19 ARDS at Mt Sinai Hospital in New York where 75% (nine of 12) patients were discharged home compared to a mortality rate of around 80% recorded in New York over the same time period in patients with COVID-19 ARDS who were not treated with remestemcel-L. Interim analyses of the 300-patient trial are planned, and Mesoblast Chief Executive Dr. Silviu Itescu is eagerly awaiting results. Curious to Know More? Age-related macular degeneration (AMD) is one of the leading causes of blindness in adults worldwide. This disease deteriorates the macula, a miraculous little spot on your retina that allows for precise vision in good light. Although several treatments exist for macular deterioration, they require frequent trips to the doctor’s office for uncomfortable but quick and routine injections directly into the eye. The required frequency of the treatments means that most patients miss appointments, leading to undertreatment of the disease and permanent vision loss. In a manufacturing collaboration with Lonza, Kodiak is designing novel antibody-biopolymer conjugate (or ABC) medicines with the same efficacy and safety with much longer durability, allowing patients to visit the doctor on a realistic schedule over the long term. By focusing on business implementation alongside formidable biotech R&D, Kodiak Sciences is on track to bring together the necessary clinical and manufacturing elements for an FDA filing in 2023. KEY TERMS: Age-related macular degeneration (AMD) is a common degenerative disease of the retina. There are two types of AMD: Dry AMD occurs when the formation of debris (drusen) on the retina causes the macula to deteriorate over time. Patients sometimes experience vision loss and frequently experience substantial functional limitations, including vision fluctuations, loss of peripheral vision, and reduced night vision. Wet AMD is an advanced form of AMD. While wet AMD represents only 10% of the number of cases of AMD overall, it is responsible for 90% of AMD-related cases of severe vision loss. Wet AMD occurs when the growth of abnormal blood vessels underneath the macula leads to leakage of fluid and blood, which leads to visual distortion, acute vision loss, and total blindness if left untreated. Vascular endothelial growth factor (VEGF) is a sub-family of factors that stimulate the growth of blood vessels. In the case of AMD, these VEGF are overexpressed, creating leaking in the macula. This leakiness causes fluid to exit from blood vessels, causing swelling – or edema – of the retina and loss of vision. An antibody biopolymer conjugate (ABC) is Kodiak Science’s proprietary platform for designing and developing drugs into the retina. The antibody in the KSI-301 molecule inhibits VEGF, while the biopolymer is comprised of phosphorylcholine, which creates a sort of “water cloak” around the antibody to increase its effectiveness. Phosphorylcholine is a natural component of the cell membrane of all the cells in our body, with remarkable properties. It attracts and binds water in a very strong – even permanent – way, creating what is known as “structured water,” which then impacts all biological interactions in the local area.

Capsule Manufacturing: It’s Not Only What’s Inside That Counts The recent EU ban on Titanium Dioxide and changing customer habits are shaking up capsule production. Over the past few years, the coloring and manufacturing of pill capsules have undergone significant changes due to new EU regulations and customer demand for natural ingredients. And while originally invented to mask and protect the contents inside a capsule, research suggests that the color of a tablet or pill can affect how patients feel about their medication. Until recently, manufacturers have primarily used Titanium Dioxide (TiO2) to create white capsules due to its efficiency in protecting the active ingredients from UV rays. However, this year an EU-wide ban on TiO2 has forced the industry to move towards alternatives that work as well, or better, than TiO2. To add to the colorant shake-up, many people are actively avoiding unnatural ingredients in their food and nutritional supplements, which has created a new demand for plant-based capsule colorants. Anticipating these changes and solving the technological challenges in a timely manner are key to a successful long-term strategy for capsule manufacturing. Curious to Know More? KEY TERMS IN CONTEXT: Pharmaceutical capsules can be either hard or soft. Soft-shelled capsules are one unique mold that encapsulates the contents, whereas the more widely-used hard-shelled capsules—such as the ones produced by Lonza—are two molded telescopic pieces of capsule: a smaller one contains the active ingredients, and a larger one encloses the capsule. Titanium dioxide (TiO2) is a widely-used pigment in capsule manufacturing, as well as in food, paint and sunscreen. Considered completely inorganic and nontoxic from a chemical point of view, it is labeled as an unnatural ingredient for ingestion, and carries the E number E171. Earlier this year, the European Food Safety Authority (EFSA) announced a six-month phasing-out ban of the colorant over concerns about nano-sized particles of TiO2 accumulating in the body. The full ban takes effect in August. The dip molding process is the manufacturing process for capsules. The final shape of the two pieces that make the capsules is defined by specifically designed molds, which are dipped in a bath of liquid formulation to pick up material that will, after the drying process, give the final capsule form, shape and composition.

Phage fermentation: Although destructive, unwanted phages can grow during fer-mentation processes for wine-making and milk production, fermentation is neverthe-less the optimal way to produce phages for therapeutic uses. Computational (science): Computer modelling of the phage and its potential inter-action with specific bacteria (known as in silico testing) allows researchers to develop phage cocktails more efficiently and with a greater chance of success.

Lab-grown meat is having a moment—the FDA just declared one company’s cultivated chicken safe to eat, and another type of lab poultry was just served for dinner to delegates at this year’s COP27 climate conference in Egypt. Whether this piques your pallet’s curiosity or turns your stomach, one thing is clear: growing meat in a lab for human consumption will take massive amounts of cell media. What is cell culture media? It is the medium in which cells grow in a lab, serving as both the cell’s food and its shelter. The medium can take various forms, depending on which cell type is being grown and for what specific purpose. It is a careful recipe that balances the complex needs of cells in nutrients, energy, pH balance and saline per-centage. Like our bodies, cell culture media is mostly water. However, crafting the right media is no simple matter: choosing the correct formula can make all the difference for the cell growth outcome, whether trying to stimulate virus production or make the tastiest animal-free cultivated cordon bleu on the planet. KEY TERMS IN CONTEXT: Cell culture media is the medium in which cells grow. It must meet all environmental conditions to keep a cell alive and flourishing. Either as a liquid or gel, synthetic or organic, the cell culture media’s most important function is to deliver nutrients to cells and to wash away waste products. The osmotic balance in cell media is the salt and water balance needed to maintain proper cell functioning. An imbalance creates uneven flows of water between a cell and its media, resulting in a cell burst or cell shrinkage. A protein factory is a name given to batches of cells cultivated in a laboratory to produce high quantities of one or several types of proteins, often for therapeutic purposes. It must not be confused with the cell’s own protein factory, aka the ribosome. The metabolomics and proteomics are the biological disciplines describing the metabolites and proteins in a cell. As these fields advance, so does the understanding of cultivating cells and developing the correct media for each type of desired growth and outcome.

While at a Big Pharma, Simon Westbrook had an idea for a complex fusion chimera protein he believed could interrupt the neurotrophin pathway and have a profound impact on chronic pain and joint degeneration in osteoarthritis patients. New Arsenal in the Battle Against Cancer: Pharmaceutical Smart Bombs Promise Less Collateral Damage for Patients Cybrexa Therapeutics CEO Per Hellsund and CSO Vishwas Paralkar talk to Lonza about how their enterprise is shaping the future of cancer treatment with cell-penetrating peptides. One of the biggest obstacles to safely eliminating cancerous cells is that most therapies also negatively impact a patient's healthy organs and tissues, known as bystanders. This notorious problem has hindered effective treatment since the beginning of oncology and often has devastating effects on patients’ quality of life. One novel strategy pursued by Cybrexa Therapeutics is the design of treatments that specifically target solid tumors made of cancer cells by taking advantage of one of their universal biomarkers – acidity. The company has developed a platform that leverages the low pH environment inherent to cancer cell metabolism. By using cell-penetrating peptides bearing an anticancer cargo load, their platform brings the treatment directly inside tumors, leaving healthy cells alone and minimizing bystander killings. This smart anti-tumor technology promises to be applicable to a wide swath of patients and reduce side effects of cancer treatment Curious to Know More? KEY TERMS: Acidity of cancer cells: Current research shows that cancer cells exhibit a type of cell metabolism known as aerobic glycolysis, a process that generates lactic acid and creates a more acidic environment in and around tumors. Cell-penetrating peptide: Peptides are strings of amino acids that can be utilized for drug delivery. They are wobbly in structure and ineffective at penetrating cells under normal pH levels but rigidify when reaching acidic environments and can then enter the targeted cell. Linkers: A chemical bond that allows for a drug to attach to its carrier and be delivered to a specific target. For Cybrexa, this bond connects the peptide to the cytotoxic molecule or DNA inhibitor for the efficient targeting of cancer cells and tumors. Cytotoxic drugs: Cytotoxic drugs in cancer therapy, such as chemotherapy, are not only toxic to cancer cells but do damage to other healthy cells, creating unwanted side effects to treatment. New research is showing how, when combined with cell-penetrating peptides, cytotoxic molecule delivery can be limited to the acidic environments of cancer cells, thereby avoiding off-target toxicity and bystander killings. DNA repair inhibitors: A relatively recent form of cancer treatment that oncologists often use in conjunction with chemotherapy. Inhibiting the repair mechanisms of cancer cells effectively turns the table on the tumor, the cancerous cells of which have hijacked the healthy DNA of a patient and use the cells natural repairing properties to become resistant to chemotherapy. By employing DNA repair inhibitors targeted specifically to cancer cells, researchers hope to increase the effectiveness of chemotherapy and reduce its side effects. Dose-limiting toxicity: When the side effects of a drug or other cancer treatment prohibit a dose increase that would otherwise be beneficial to the cancer therapy. By removing this boundary, therapies could be much more efficient in destroying cancerous cells in the body.

De-risking Drug Development: Early Testing for Toxicity Saves Time and Resources Yvette Stallwood, head of Lonza’s Early Development Services, talks about patient safety and other advantages of early testing for immunogenicity in the drug development pipeline. In the high-stakes drug discovery game, from IND filings all the way up through the clinical trial phase, regulatory authorities are now expecting developers to have an immunogenicity risk strategy in place. “It really is essential that drug developers assess the immunogenicity risk as early as possible in the pipeline, as not only can it impact the functionality of the drug, but it can also be a significant safety risk for the patient,” explains Yvette Stallwood, whose work at Lonza’s Early Development Services (EDS) is helping small and large biotech companies reduce risk with a “Right First Time” approach when developing drug therapies. Drug candidates often fail during clinical trials due to their toxicity to patients, which is evident, for example, in an immunogenic reaction—where the drug triggers an unwanted immune response known as immunotoxicity. This can result in the loss of years of work and funding. Stallwood and her team encourage their clients to begin with in-silico testing, where up to a thousand digital models of potential immunoresponses can be predicted. Once the digital models show a candidate to have a low risk of toxicity, the EDS team then moves to human donor cell assays—with the advantage of screening up to fifty different immunotypes. The ideal time to assess immunosafety and immunotoxicity is well before deciding on a molecule as a lead drug candidate. By understanding as much as possible about the potential product through early testing, biotech companies are better equipped to take the correct path to regulatory approval with a drug that is ultimately safer for patients. KEY TERMS IN CONTEXT: Anti-drug antibody (ADA) response happens when the patient’s immune system generates antibodies to remove and clear the drug from the body. This can impact the effectiveness of the drug molecule as well as be dangerous for the patient. Immunogenicity testing is the process by which one can test for the body’s immune response to a drug. With in-silico testing, the screening can be done quickly for a large swath of different immune system typologies before moving on to animal models or, preferably, human cell assays. Immunosafety and immunotoxicity refer to how potentially safe or toxic the immune system’s reaction to a molecule may be. They deserve the utmost consideration when developing a leading drug candidate. De-risking is an EDS drug development strategy to ensure clients select the right drug candidate at the approval phase. De-risking avoids costly clinical trial failure through extensive immunotoxicity testing early in the process. In-silico testing uses computer models to test a molecule’s reaction within an organism, such as a human immune system. The advantage is that they are quick and can test in hundreds and thousands of models. Since they are limited in their nature, they are only a first step. Once a drug candidate is selected as low risk using in-silico testing, further testing is needed using animal models or human cells assays. Human cell assays, in the context of drug development de-risking, are in-vitro tests that use actual human immune cells to test immune system responses to drug candidates. Although they are more costly and time-consuming than in-silico testing, the precision they offer is essential to establish the appropriate data for selecting lead drug candidates.

Corvus is a small biotech developing precision medicines, primarily for oncology patients. It came to Lonza seeking to develop a monoclonal antibody called CPI06, now known as Mupadolimab. This inhibits the production of adenosine in the tumor microenvironment and has an agonistic effect on B cells. When considering CDMOs for CPI06, Corvus was aware of Lonza’s platform methodology, custom built for mAbs, and its track record commercializing antibodies. Lonza welcomed Corvus and demonstrated flexibility taking on the project by starting work with available capacity in Singapore and then transferring to Slough, UK. Multiple consistent batches were manufactured, and the product is currently in clinical trials with CMC work simultaneously taking place to provide a good regulatory platform with a goal for commercialization.
